| | | calibration article ? any one care to start off with a nice and easy to follow " how to calibrate the microscope graticule" article ?
ive had a go with mine, but the spores i tried to measure (from Agaricus sainsburyii) did not seem to fit in the given ranges, making me suspect that i have not got the calibration right,
with emphasis on the "easy to follow" format for microscope dummies like me
great to see the introduction of this sub-forum, by the way,
it should help open a few doors for a better understanding of the subject,

If so, for each different magnification lens work out how many units on your eyepiece graticule equate to a fixed number of microns on the stage micrometer. You can then work out the ratio. i.e. if at 10x magnification 100um on the stage micrometer matches with 130 units on your eye piece graticule then you know that 1 unit on your eye graticule =100/130 um. So that is the ratio you need to multiply all your measurements by to get it into microns (um). You need to do this for all the magnifications as 4x lens ratio will not necessarily be 10 times the 40x lens ratio.
As it happened, with my microscope 4x, 10x, 40x was spot on. I needn't have checked it and calibrated it with the stage micrometer. But I didn't know it was that accurate until I checked it
That is probably as clear as mud ...

I'm not sure how effective the video will prove, because I have java switched off on my computer.
However, there are (written) descriptive articles on this site as well, which are very comprehensive.

i used something similar to Melanies method, but only on the 10x thinking it would be the same for the other settings (20x & 40x) and i used the 40x for the actual measurement, so in all probability it was way out, which would explain the dubious results i got,
ive had a quick look at Mikes suggested website and by using that as a guide i think i will be able to make a more accurate job of it the next time
a quick trip out for some more Agaricus sainsburyii
